Clarke was writing the novel all along. But the movie script had to freeze long before the book, and in the interim, astronomers managed to rule out parts of the script. Clarke is a hard SF writer so he moved the book setting from Jupiter to Saturn get around the problem. The result of the tinkering was significant differences between book and movie.

Clarke generally didn't do sequels - I can't think of any until much later - and of course Kubrick never did them. I don't think they ever considered sequels at all.
