I'm pretty sure it's something to do with the slate/shot number, but if anyone could shed any extra light on it (like someone who has worked on one of PT's sets, for example), I would be most grateful.
Actually, DanG is correct. It is the scene number said during the slate. In his first example, the script supervisor would know that it was Scene 10A, Take 1. The second shot setup in Scene 10 would be Scene 10B. Once you run out of the alphabet sequence, you go to 10AA.
A scene is generally understood as a single location at a certain time in the script. If in the movie, the story sequentially goes from day to night, but is the same location, that would be considered the next scene. But simply changing the camera angle keeps the same scene number with a new letter.
